Chelating Agents for Radioisotopes

Chelating agents such as DOTA, DOTEM and TCMC are often used to radiolabel targeting molecules for cancer therapy. These common chelating agents can bind beta-emitting radioisotopes well enough to deliver the radiation to the targeted cancer cells but they are much less effective at binding more potent alpha-emitting radioisotopes. When an alpha-particle is emitted, the “re-coil effect” can dislodge the radioisotope from the chelator and cause off-target toxicity.
